Research Summary
As a principal investigator, I lead a lipid research group in UCL, Royal Free Campus. This group is the only dedicated renal-oriented lipid research group in the UK and is probably the only one with a long track record and that is equipped to carry out such studies. My major goal has been to develop an academic research programme that investigates the involvement of lipoproteins in renal and vascular diseases to address clinical problems associated with progression of renal disease, chronic allograft dysfunction, cardiovascular disease associated with renal failure and the link between inflammation and lipids in the development of atherosclerosis. My current focus is on the influence of inflammatory stress on cellular cholesterol homeostasis. We have demonstrated that inflammatory stress results in lipid accumulation in peripheral tissues (cells) by disrupting cholesterol trafficking and causing cholesterol re-distribution. The works so far have given us a solid basis for the further exploration of the influence of inflammation in aggravating lipid-mediated vascular and renal injury. The study of the response of intracellular cholesterol homeostatic mechanisms to inflammatory stress may eventually yield a 'wiring diagram' that links several pathways (lipoprotein receptors and ABCA1, etc) together via molecular 'cholesterol sensors' (e.g. SCAP) and 'switches' acting in sequence. I believe the consistent acceptance of our papers by high impact factor journals is a good validation and encouragement for future work in this area. I have been especially encouraged by the enthusiasm of established researchers worldwide to collaborate with me.
Teaching Summary
My teaching at UCL involves the intercalated BSc in Molecular Medicine (the half course unit in cellular and molecular aspects of cardiovascular disease) and Biochemistry C41-C43 (Biochemistry in Health and Disease) for undergraduates. These courses are designed to take advantage of new developments in molecular and cellular biology and their applications to clinical medicine. The students obtain ?hands on? experience of the latest laboratory techniques and their applications to clinical and basic research problems in molecular medicine (genetics, cardiovascular, molecular endocrinology, reproduction and oncology). The course provides the basis of the ?molecules to disease? approach of the future. My lectures are aimed at giving the students an understanding of the molecular basis of renal dysfunction and cardiovascular disease. In addition, I supervise the research projects of one or two BSc students per year for the fulfilment of their intercalated BSc course in the renal research unit in the Centre for Nephrology.
