Cheng is a team member of EPSRC funding "Enabling rapid liquid and freeze-dried formulation design for the manufacture and delivery of novel biopharmaceuticals". He undertakes an Innovate UK secondment program for Botulinum toxin (BoNT) with Ipsen. He is a Member of Royal Society of Chemistry (MRSC).
Protein Engineering and Formulation Optimisation
- Computational modelling for the Botulinum toxin (BoNT), Targeted Secretion Inhibitors (TSI) and single-domain antibodies (VHH)
- Computational modelling for the A33 Fab, including Rosetta and molecular dynamics (Gromacs)
- In silico identify the unstable residues of Fab and propose stabilising mutations
- In vitro express and purify the Fab variants
- Validate the variants’ stability by melting temperature (Tm) and aggregation kinetics
- Stabilise the Fab through increased concentration (DLS, SLS, viscosity, crowding agents)
Crystallisation
- Screen out the potential crystallisation conditions for Fab
- Resolve the X-ray structure and deposit to PDB (7NFA, 7NC0)
- Determine the structural alteration in Fab variants and formulation conditions
Freeze-drying
- Coordinate a freeze-drying training course in UCL with Biopharma company
- Develop a freeze-dried dosage form for the Oxford-AstraZeneca ChAdOx1 COVID-19 vaccine
