The synthesis and investigation of chimeric luciferins
There is a pressing need for bright, multicoloured, nr-IR emitting luciferins for improved in vivo tissue penetration and new multiparametric bioluminescence analytical/imaging techniques for a wide range of applications.The purpose of this project is to answer the fundamental question of whether or not chimeric luciferins can bioluminesce. The design of the chimeric luciferins will follow from careful consideration of the chemical reactivity of individual luciferins and the promiscuity of luciferases. The project will combine crucial molecular features of luciferins from two different natural systems to make novel unnatural chimeric luciferins. We will explore tuning for nr-IR multiparametric bioluminescence imaging for those that exhibit bioluminescence and show superior characteristics to natural luciferins.
