Research Summary
I have worked for about 17 years on different aspects of regulation of gene expression by changes in oxygen - sparked by an initial interest in how the kidney and liver regulate production of the hormone erythropoietin (EPO). Early discoveries while working as an MRC Clinical Training Fellow in Peter Ratcliffe's lab in Oxford included identifying the EPO producing in the liver and kidney, and showing that the underlying oxygen sensing system appeared to operate in all mammalian cells. Considerable progress has been made since; the system involves activation of a transcription factor Hypoxia Inducible Factor-1 (HIF) and controls processes ranging from angiogenesis to cellular metabolism. Key discoveries included (1) establishing the role of the von Hippel Lindau (VHL) tumour suppressor gene in HIF alpha destruction (2) that prolyl hydroxylation provided the recognition signal for capture by VHL (3) identification of the PHD enzymes that hydroxylate HIF alpha subunits, acting as oxygen sensors. With others we have shown that congenital polycythaemia (erythrocytosis) in humans can be caused by homozygosity for a subtle defect in VHL, a heterozygous mutation in PHD2 or an activating mutation of HIF-2alpha. The current program is focused on the link between VHL loss and kidney cancer, and the therapeutic potential of potentiating HIF activation by inhibiting the hydroxylase enzymes. We have now generated mice lacking each of the three PHD enzymes which hydroxylate HIF, and shown that lack of PHD1 is protective from ischemic injury.
